Screening of Various Organic Substrates and the Development of a Suitable Low-Cost Fermentation Medium for the Production of a-Amylase by Bacillus subtilis

چکیده
The production of extracellular amylase by Bacillus subtilis has been studied in solid-state fermentation (SSF). In a sequential order, various process parameters were optimized for maximum amylase production. The tested process parameters were different solid substrates such as banana husk (BH), water melon husk (WMH), lentil bran (LB), wheat bran (WB), melon husk (MH) and maize oil cake (MOC), different incubation time (24–144 h), particle size (500–2500 mm), initial moisture content of the substrate (40–70 %, by mass per volume), inoculum size (10–60 %, by mass per volume) and inoculum concentration (10–60%, by volume per mass). The maximum production of amylase (4857 U/mg) was achieved when 1 % starch (by mass, particle size of 1500 mm) was added to banana husk as the solid substrate, fermented for 72 h at 37 °C, at an inoculum level of 30 % (by volume per mass), and initial moisture content of 60 % (by volume per mass).

Evaluation of Ca-Independent a-Amylase Production by Bacillus sp. KR-8104 in Submerged and Solid State Fermentation Systems
This study investigates the production of crude Ca-independent and low pH active α-amylase by Bacillussp. KR-8104 in submerged fermentation (SmF) and solid-state fermentation (SSF) systems. Differentparameters were evaluated in each system using “one factor at a time” approach to improve the production ofenzyme. The results showed that in the SmF the maximum enzyme production ...

A study was carried out with a newly isolated bacterial strain yielding extracellular amylase. The phylogenetic tree constructed on the basis of 16S rDNA gene sequences revealed this strain as clustered with the closest members of Bacillus sp. and identified as Bacillus subtilis BI19. The effect of various fermentation conditions on amylase production through shake-flask culture was investigate...
متن کاملAmylase production from Aspergillus oryzae LS1 by solid-state fermentation and its use for the hydrolysis of wheat flour
Nine Aspergillus and three of Trichoderma strains were grown on wheat bran (WB) medium under solid state fermentation (SSF) for amylase production. Aspergillus oryzae LS1 produced the highest level of the enzyme. The thermal stability profile of its crude enzyme revealed the half-life time of more than 2 h at 50 and 60ºC. The enzyme production was affected by strain type, incubation periods, le...
متن کاملSignificantly enhancing recombinant alkaline amylase production in Bacillus subtilis by integration of a novel mutagenesis-screening strategy with systems-level fermentation optimization
BACKGROUND Alkaline amylase has significant potential for applications in the textile, paper and detergent industries, however, low yield of which cannot meet the requirement of industrial application. In this work, a novel ARTP mutagenesis-screening method and fermentation optimization strategies were used to significantly improve the expression level of recombinant alkaline amylase in B. subt...
متن کاملResponse of surface optimization for the enhanced production of alkaline protease isolated from Bacillus sp. with bean husk as a new substrate
Optimization of the fermentation medium for maximum alkaline protease production was carried out. Fifteen positive isolates were examined for their extent of alkaline protease production. The most potent producer was identified as Bacillus sp. The solid substrate screening showed that the combination of wheat straw and bean husk was the best one. The initial screening by using Plackett–Burman’s...
